Description:
GCaMPisageneticallyencodedcalciumindicator(GECI)andcreatedfromafusionofgreenfluorescentprotein(GFP),calmodulin(CaM),andM13,apeptidesequencefrommyosinlightchainkinase. WhenGECIbindsCa2+,theconformationalchangeofGCaMP inducesproximityofthe GFPparts,eliciting strongGFPfluorescencesignal,whichallowsmeasuringactionpotentialsandotherreceptoractivationeventsthattriggerCa2+influx.TheadvantageofGECI"sarethattheycanbegeneticallyspecifiedforstudiesinlivingorganisms.
GCaMP6isanovelultra-sensitiveformatofGCaMPthatoutperformedothersensorsintermsofaccuracyandsensitivityinmeasuringcytosolCa2+level. MutationsincalmodulinpartofGCaMP6createdseveralvariants whichshoweddifferentcalciumfluorescencedecayrate(fast- GCaMP6m, slow -GCaMP6m andmedium-GCaMP6m).
TheAAV5-CaMKII-GCaMP6misapre-packagedrAAVinSEROtype5(withcapsidfromAAVserotype5and2xITRfromAAVserotype2)whichover-expressesgeneticallyencodedcalciumindicatorGCaMP6m underCaMKIIpromoterforneuronexpression.
